As shown in the illustration, the temporal and tissue-specific expression pattern of a gene is determined by its particular set of enhancers. Enhancers are recognition sites for activator proteins. These proteins stimulate transcription by physically interacting with RNA polymerase and other general transcriptional factors bound at the promoter. Because enhancers are often far removed from their promoters, this interaction requires looping of the DNA.
